This is all for repentance, according to the most recent version of BoI. In this section, I’ll provide a brief summary before devoting one section to each character, including their tainted variant.
Going off the completion mark sheet in the game, generally left to right then top to bottom, formatted as follows:

Brimstone Symbol (Mega Satan) Ch6 bonus boss (find 2 angel rooms, bomb the angel statues and beat the angels for both parts of the key, go up with polaroid or down with negative to reach floor 6. Alternatively for key, use sacrifice rooms to summon angels):

Star (Boss Rush) Beat Mom (Ch3 fl2) within 20 minutes, 25 minutes if you go the alternate route. Alternatively, have Broken Shovel or Mama Mega active and it will open regardless of time. There is also an exploit with Berserk! and Suplex!, but I haven’t done it before:

Knife (Mother) 2nd Boss of The Corpse, Follow the alternate path to get the knife pieces, sacrifice the knife to open the flesh door at the end of Mausoleum/Gehenna 2, alternatively, sharp key can be activated to open it, Cracked orb can be triggered, Soul of Cain can be used, or sometimes exiting the boss rush room after 25 minutes have passed will move the door in a way that leaves it open:

Dad’s Note (The Beast) Follow the Ascent Path to get to Home (unlocked by defeating Mother) and beat both Dogma and The Beast, you should look this one up, it’s also connected to unlocking tainted characters.
